A New Dimension of Immune Checkpoints Interactions

T cell mediated tumor destruction is restricted by trans-interactions between immune checkpoint receptors and their ligands. Checkpoint inhibitors have demonstrated impressive clinical activities against some human cancers, but only in a small subset of patients. I will discuss our recent findings that in addition to trans-interactions, checkpoint receptors and ligands can interact in cis on the same cells, and that how these cis-interactions can profoundly impact the anti-tumor activities of T cells and immune checkpoint inhibitors.
